Only seven names, and two in the manga. The Twelve Guardian Ninja, an elite guard in the exclusive service of the Daimyo of the Land of Fire, come mostly from the databook and the anime. The manga shows only two of them, Asuma Sarutobi and Chiriku, met during the Hidan and Kakuzu arc; the other five all come from the big Shippuden filler arc. Sorting canon from filler, the group's distinctive mark, the chakra blades, and Asuma's falling-out with his father.
A lord's elite guard, and the secret behind Asuma's falling-out with his father: the Twelve Guardian Ninja are one of those groups the manga only touches on and the anime fully unfolds. Let's untangle canon from filler.

The Fire Daimyo's Guard
The Twelve Guardian Ninja are an elite guard of twelve shinobi in the exclusive service of the Daimyo, the feudal lord of the Land of Fire. Their membership is signaled by a cloth worn at the waist, bearing the kanji for "Fire".
The concept appears in the manga in passing, during the arc hunting down the Akatsuki, when a character reveals he was once part of it. Former Guardians keep a reputation — and bounties — in criminal circles.

Asuma, Konoha's Guardian
That character is Asuma Sarutobi, the son of the Third Hokage Hiruzen. He reveals that he and the monk Chiriku both belonged to the Guardians.
According to the databook, Asuma is said to have left Konoha years earlier, after a disagreement with his father over the shinobi life, and to have become a Guardian during that absence — hence the distance between them.

The Chakra Blades
The most famous weapon associated with the group is in fact Asuma's signature: trench knives worn like brass knuckles, made of a metal that can be infused with a chakra nature.
Through his Flying Swallow technique, Asuma channels Wind Release chakra into them to extend their reach and sharpen the blade. Shikamaru will inherit them after him. Note: nothing establishes that all twelve wielded them.

Only Two Members in the Manga
This is the limit to know: in manga canon, only two members are identified — Asuma and Chiriku. Seven names exist in total, but the other five all come from filler.
So it is impossible to reconstruct a complete "roster" of the twelve: most of them remain anonymous. What the duo Hidan-Kakuzu is hunting is precisely the bounties attached to these former Guardians.

Sora's Filler Arc
All the group's detailed development belongs to a filler arc (the first one in Shippuden), built around the young monk Sora, a Wind Release chakra user like Naruto.
Sora is presented there as a "pseudo-jinchuriki": he carries residual chakra from the Nine-Tails, which escaped during the attack on Konoha, not the beast itself. Sora, the coup and the reanimated Guardians are original anime, not manga.

FAQ
-
Are the Twelve Guardian Ninja manga or filler? Both, but in very unequal doses. The concept and two members (Asuma, Chiriku) are manga canon: Asuma reveals it during the arc hunting down the Akatsuki. All the detailed development — Sora, Kazuma, the reanimated Guardians, the coup — is a filler arc of Naruto Shippuden, the first one in the series.
-
Why had Asuma distanced himself from his father Hiruzen?According to the databook, Asuma left Konoha years before the start of the story after a disagreement with his father over the shinobi life, and joined the Daimyo's Guardians during that absence. This point comes mainly from the databook and the anime; the manga does not detail it.
-
How many of the twelve members are actually named?Seven in all. In manga canon, only Asuma and Chiriku. Filler adds five names: Kitane, Nauma, To and Seito (the loyal Guardians who are reanimated) as well as Kazuma (the leader of the plot). The others remain anonymous: a complete roster of the twelve should not be reconstructed.
-
Is Sora a real jinchuriki of the Nine-Tails?No. Sora is a "pseudo-jinchuriki" specific to filler: residual Nine-Tails chakra, which escaped during the attack on Konoha, was collected and then sealed inside him. So he carries a share of the Nine-Tailed Fox's chakra, not the tailed beast itself (sealed inside Naruto). He is an anime-exclusive character.
-
What are Asuma's "chakra blades"?They are his trench knives, worn like brass knuckles, made of a metal that can be infused with a chakra nature. With his Flying Swallow technique, he channels Wind Release chakra into them to extend their reach and sharpen the blade. It is Asuma's signature — not a weapon shared by all twelve — and Shikamaru inherits them after his death.
In summary
The Twelve Guardian Ninja are a fine example of how Naruto works: an idea dropped in a single line by Asuma, which the anime turns into an entire arc. Keep the canon essentials — an elite guard, two members, and the key to the relationship between Asuma and his father.
